Molecular characterisation of a meropenem-resistant Haemophilus influenzae

چکیده

Background: Haemophilus influenzae β-lactam resistance is mediated by production of β-lactamase and mutations in penicillin-binding protein 3 (PBP3, encoded ftsI). Only one the 646 patient-unique H. isolated our laboratory between 2012–July 2022 was meropenem (amoxicillin, co-amoxiclav ceftriaxone) resistant. Methods: Identification susceptibility this meropenem-resistant (MRHI) confirmed disk diffusion E-test (bioMérieux) (CLSI M100). WGS performed on GridION (Nanopore) reference mapped with Geneious Prime 2022.2. PCR Sanger sequencing used to confirm results WGS. Results: The MRHI isolate negative non-typeable. ftsI subgroup IIc1 were present, signifying previously described low level PBP3 alterations that should not solely account for carbapenem resistance. Group VII2 acrR (repressor AcrAB efflux pump) substitutions observed but without reported associated frameshift mutation. There also multiple insertions ompP2 (outer membrane P2) porin, resistance.2 100% concordance Nanopore increases confidence application technology future investigations. Conclusion: Meropenem likely multifactorial, decreased influx as contributing factors. References 1. Dabernat H, et al. Diversity resistance-conferring amino acid influenzae. Characterization of chloramphenicol-resistant Haemophilus influenzae.

We examined nine chloramphenicol-resistant (minimal inhibitory concentration, greater than or equal to 15 micrograms/ml) Haemophilus influenzae strains isolated in various parts of the world to characterize the genetic and biochemical bases of the resistance; four were type b. All nine contained conjugative plasmids, ranging in molecular weight from 34 x 10(6) to 46 x 10(6), which encoded for r...

Levofloxacin-Resistant Haemophilus influenzae, Taiwan, 2004–2010

Levofloxacin resistance in Haemophilus influenzae has increased significantly in Taiwan, from 2.0% in 2004 to 24.3% in 2010 (p<0.001). Clinical and molecular investigations of 182 levofloxacin-resistant isolates revealed that the increase was mainly the result of the spread of several clones in the elderly population in different regions.

Haemophilus Influenzae B

Vaccine Candidates against Nontypeable Haemophilus influenzae: a Review

Nonencapsulated, nontypeable Hemophilus influenzae (NTHi) remains an important cause of acute otitis and respiratory diseases in children and adults. NTHi bacteria are one of the major causes of respiratory tract infections, including acute otitis media, cystic fibrosis, and community-acquired pneumonia among children, especially in developing countries. The bacteria can also cause chronic dise...

Quinolone-resistant Haemophilus influenzae in a long-term care facility: clinical and molecular epidemiology.

We describe a clonal outbreak of quinolone-resistant Haemophilus influenzae (QRHI) from an affiliated long-term care facility (LTCF-A); the outbreak was associated with the clinical use of levofloxacin, which was determined to be a risk factor for acquisition of QRHI. The minimum inhibitory concentration to which 90% of isolates were susceptible (MIC90), as determined by broth microdilution, wa...
